The fictional Melbourne neighborhood of Erinsborough will welcome her as a guest star, playing the role of Reece, according to a Monday announcement from production company Fremantle and streaming service Amazon Freevee.
Reece, an American who is new to the region, will "not quite be who she appears to be," according to the show's producers.
According to a press release from the executive producer Jason Herbison, they are "beyond excited to have an actor of Mischa's caliber join us for the beginning of this exciting new chapter."
"Mischa's character is unexpected and vibrant, and he or she will be immediately recognizable on Ramsay Street. Both domestically and internationally, our devoted viewers will adore her, he continued.
The popular drama series, which aired for nearly 40 years, came to an end in July.
The revival series will enter a new chapter with the upcoming season, which began filming in Melbourne on Monday and will make its debut in the fall.
In the statement, Barton said, "I'm thrilled to be a part of this legendary show's next chapter, and I am particularly looking forward to being back in Australia - a home I know and love.
When Barton was young, she made her acting debut in the late 1990s films "Notting Hill" and "The Sixth Sense."
The star soon became well-known for her on-screen depiction of the title character Marissa Cooper in "The O.C." from 2003 to 2006.
Together with actor Bruce Willis, she starred in the 2008 crime movie "Assassination of a High School President" and the 2009 suspense picture "Homecoming."
Barton now joins celebrities like singer Kylie Minogue, actress and "Barbie" star Margot Robbie, and actor Guy Pearce who have all made appearances in "Neighbours."
According to the show's creators, Tim Kano (as Leo Tanaka), Rebekah Elmaloglou (as Terese Willis), Georgie Stone (as Mackenzie Hargreaves), and Annie Jones (as Jane Harris) will all reprise their respective roles.
Additionally reprising their roles are special guests Ian Smith (as Harold Bishop), April Rose Pengilly (as Chloe Brennan), and Melissa Bell (as Lucy Robinson).
